Topical oxygen therap combined with vacuum-assisted closure intervention wound healing of III period pressure ulcer in the left trochanter major of guinea pig

Objective:To investigate the Effect of topical oxygen therap combined vacuum-assisted closure intervention on the treatment of III period pressure ulcer in the left trochanter major of guinea pig,and to optimize working parameters.Methods:The experiments included comparative efficacy and parameter optimization.Comparative efficacy of experiment: selected 20 clean male white guinea pig,construction of III period pressure ulcers in the left trochanter major of guinea pig.20 wounds were divided randomly into five groups: Group A were treated with Continuous VAC(-125mmHg) for 320 min and TOT(5L/min) for 160min;Group B were treated with VAC(-125mmHg) for 40 min and TOT(5L/min) for 20min,Alternating 8times;Group C were treated with VAC(-125mmHg) for 40 min after suspension of 20min,Repeat 8 times;Group D were treated with TOT(5L/min) for 20min after suspension of 40min,Repeat 8 times;Group E were treated with conventional double saline gauze and sterile porous polyurethane sponge dressing change every other day.Measuring wound area at the given time point(immediately the wounds were created and the 1 st,3rd,7th,11th day after the treatment),then calculate wound shinkage rate.Parameter optimization of experiment:construction of III period pressure ulcers in the left trochanter major of 23 guinea pig.The uniform design and multiple regression analysis were applied to investigate topical oxygen therap combined with vacuum-assisted closure intervention wound of III period pressure ulcer healing for intensity of negative pressure,oxygen flow,within an hour topical oxygen therap working time,intervention frequency on daily average of wound shrinkage rate.Results:The wound shinkage rate at the given time point of group A and B were higher than that of other grouds(P0.001).The best working parameters for topical oxygen therap combined vacuum-assisted closure intervention in guinea pig model of III period pressure ulcer was ascertained as follows: intensity of negative pressure=-125mmHg,oxygen flow= 10L/min,within an hour the TOT working time=50min,intervention frequency=12 times / day.Conclusion:VAC and TOT combined intervention therapy can significantly promote wound healing of III period pressure ulcer in guinea pig.The uniform design and regression model can be used to precisely predict the test results,and optimize the working parameters.
