•1,25-dihydroxyvitamin D3 reduces melanoma cell viability while increasing caspase levels.•1,25-dihydroxyvitamin D3 inhibits PI3K-AKT-mTOR and RAS-ERK signaling pathways in melanoma.•The growth inhibitory effect of 1,25-dihydroxyvitamin D3 in melanoma cell lines is dependent on PTEN and the vitamin D receptor (VDR).•1,25-dihydroxyvitamin D3 may target PTEN to reduce melanoma cell viability.•The role for vitamin D in melanoma remains unclear but it may have therapeutic benefit.
