•6-OHDA taken up through dopamine transporters produces H2O2 in dopaminergic neurons.•H2O2 is retrogradely transported to presynaptic glutamatergic terminals.•H2O2 activates TRPM2 channels and glutamate accumulates in the extracellular compartment.•H2O2 induces intracellular Zn2+ dysregulation via AMPA receptor activation.•Zn2+ dysregulation results in dopaminergic degeneration prior to movement disorder.
