Optimization of experimental variables for the production of-amylase by aspergillus oryzae using rice bran

This investigation is made to optimize the experimental parameters for the production of amylase by Aspergillus oryzae MTCC-8624 on using rice bran as a cheaper substrate. pH, temperature, fermentation time, inoculum concentration and substrate using a Box–Behnken design under the response surface methodology. The optimum values of experimental parameters are found to be pH4.7; Temperature 36.5C; Fermentation time 66.6hr; Inoculum concentration 5.7% and Substrate concentration 18.9 g/L. At these optimized conditions maximum amylase activity is found to be 16.89 U/ml.
