Effects Of The Interaction Between Mir-1466-5p And Ccdc6 On The Biological Behavior Of Papillary Thyroid Carcinoma Tpc-1 Cells

Objective: To study the effects of mocroRNA-146b-5p (miR-146b-5p), coiled-coil domain-containing-6 (CCDC6), and their interation on the biological behavior of papillary thyroid carcinoma TPC-1 cells.Methods: This study selected 25 human papillary thyroud carcinoma tissue samples and 25 samples of normal thyroid tissue adjacent to cancer tissues stores in the study hospital between 2015 and 2017. The relative expression levels of miR-146b-5p and CCDC6 for each tissue sample were determined using qRT-PCR. The papillary thyroid cancer cells were into a miR-NCgroup (miR-146b-5p non-specific control lentiviral vector), a miR-146B-5p-inhibitor group (constructed lentivirus and down-regulated miR-146B-5p expression), a miR-146b-5p group (constructed lentivirus and up-regulated miR-146B-5p expression), aLv-miR-146b-5p + Lv-CCDC6 group (constructed lentivirus and down-regulated miR-146B-5p; up-regulated miR-146B-5p expression), a si-NC group (CCDC6 non-specific control vector), and a si-CCDC6 group (constructed lentivirus and down-regulated CCDC6 expression). At 0 H, and 24 h, the study compared CCDC6 protein expression in the miR-146b-5p-inhibitor groups and the proliferation, invastion ability of the other groups.Results: The relative expressions of miR-146b-5p was significantly higher in cancer tissues than that in normal cancer-adjacent tissues; the relative expression of CCDC6 was siginificantly lower in cancer tissues than in normal cncer-adjacent (P<0.01). The CCDC6 expression levels for the miR-146b-5p-inhibitor + Lv-CCDC6 groups was significantly lower han that of the miR-146b-5p group (P<0.05). At 24 h, the cell migrations ability of the si-CCDC6 group wassignificantly higher than that of the si-NC group (p<0.05). At 12 h and 24 h, the average number of transmembrance cells in the Lv-miR-146b-5p _ Lv+CCDC6 group wassignificantly lower than the number in the miR-146b-5p group (P<0.05). At 24 h, the average number of transmembrane cells in the si-CCD6 group was significantly higher than the number in the si-NC group (P<0.05).Conclusion: Up-regulated CCDC6 expressions and inhibited miR-146B-5p expressions can hinder the proliferation, migration, and invastion of papillary carcinoma TPC-1 cells, amomg other biological behaviours,
