Development and characterization of 36 SNP markers for Hynobius yiwuensis

The population of Hynobius yiwuensis has decreased dramatically due to urbanization and human activities. In order to better monitor and protect this species, 36 single nucleotide polymorphism (SNP) markers were developed using double digest restriction-site associated DNA sequencing (ddRAD-seq). The frequency of minor allele per locus ranged from 0.0132 to 0.3594, while the observed and expected heterozygosity varied from 0.0263 to 0.5278 (average 0.1565) and from 0.0260 to 0.4604 (average 0.1396), respectively. The inbreeding coefficient ranged from − 0.2643 to 0.0000. No loci showed significant departure from Hardy–Weinberg equilibrium ( P HWE > 0.05). These polymorphic SNP markers will be useful for the study of conservation genetics in H. yiwuensis .
