•Glucopyranosylchrysin isolated from Mimosa rosei was derivatized and studied by VCD.•A VCD methodology for the AC determination of a glycosylflavonoid is described.•The VCD statistical validation of a glucopyranoside is described for the first time.•The vibrational mode values for glucopyranosylchrysin peracetate were characterized.•Immunomodulatory response of glucopyranosylchrysin and its peracetate was evaluated.
