Rutin Extraction And Quantification From Sophora Japonica L. And Antioxidant Capacity Analysis

In this paper, various extraction methods and quantification of rutin from Sophora japonica L. were developed and antioxidant capacity of rutin was assayed by different measurements . The performance of extraction can be evaluated based on the k(d) (distribution coefficient), which reflected the migration and separation capacity of the solute. The quantification was carried out by calculation formula of extraction efficiency with solvents. Based on the method, rutin content in Flos Sophorae buds from different regions was quantified and the highest content of rutin was more than 22% of dry weight. Rutin content in Flos Sophorae buds was higher than that of flowers and that in flowers of initial flowering stage was higher than that in flowers of full flowering stage, which provided optimum acquisition time. In addition, rutin possessed higher antioxidant activity and free radical scavenging activity in vitro experiment. DPPH radical scavenging activity was 2.2 +/- 0.02 times of vitamin C equivalents (mass ratio), and ABTS radical cation scavenging activity was 2.38 +/- 0.075 time of trolox equivalents (mass ratio), and reducing powers was 1.79 +/- 0.070 times of vitamin C equivalents (mass ratio), which enable us to see the probable function of rutin in plant cells.
