摘要
Collagen polypeptide was prepared from cattle hide by two-step method with proteinases. Reaction mechanism between collagen polypeptide and calcium was characterized with FTIR spectra and the method of comparing the reaction abilities with calcium among collagen polypeptide and modified collagen polypeptides. By combined with appropriate calcium compound, a new calcium supplement preparation (CPCAG) was made which supplied not only calcium but also collagen. The beneficial effects have been proved by application experiments with mice. A mice model of osteoporosis was obtained with administering retinoic acid 70mg/kg per day by gastrogavage for two weeks. Three feeds containing different calcium source were given to osteoporotic mice. After 20 days and 40 days treatment, the mice were sacrificed separately, and some blood biochemical parameters and bone parameters such as alkaline phosphatase (ALP) activity, serum calcium concentration, bone mineral content (BMC) and bone hydroxyproline (HYP) concentration were measured to verify the beneficial effect of CPCAG. In addition, scanning electron microscopy (SEM) was used for observing changes on mice bone structure. The results manifest that the effect of this new calcium supplement preparation was satisfied, and it does support the suggestion that collagen hydrolysate plays a positive role in assisting the absorption of calcium.
