Interleukin 24 Inhibits Growth And Induces Apoptosis Of Human Large Cell Lung Cancer Cell Line Nci-H460 Transplantation Tumor In Vivo

Objective : To study the enhanced anticancer effect and its mechanism by Ad-polyA-promoer-IL-24 (Ad-IL-24) for human large cell lung cancer cell line NCI-H460 transplantation tumor. Methods : Anti-tumor experiment was conducted on these mice bearing lung adenocarcinoma transplantation of NCI-H460 cell by intratumorally injecting with the recombinant adenovirus. And tumor growth was recorded by volume. All tumors were taken out and weighed at three days after treatment completed. The expressions of cytokines including Bax, Caspase-3, Bcl-2 and Survivin were detected by immunohistochemisty. Results: Tumor growth was significantly inhibited in the Ad-IL-24 group as compared with that in the Ad-GFP group and the PBS group (P<0.05). The weight inhibitory rate was 32.69% in the Ad-IL-24 group. The expressions of Bax and Caspase-3 were up-regulated, and the expressions of Bcl-2 and Survivin were downregulated in the Ad-GFP group. Conclusion: It is proved that there is enhanced anticancer effect by Ad-IL-24. The mechanism may be related to the up-regulations of Bax, Caspase-3 and the down-regulations of Bcl-2, Survivin.
