Effect Of Levo-Anlodipine On Myocardial Cells Calcium Overload In Anoxic Conditions

Objective: To observe the influence of levo-Anlodipine to calcium overload of the anoxic myocardial cells. Methods: By the cultivation of myocardial cells in vitro and the establishment of anoxic myocardial cells, we used Fura2/am fluorescent probe to measure the free calcium concentration of the myocardial cells through spectrofluorometer directly and assayed Ca(2+) ATPase of cytomembrane and sarcoplasmic reticulum through RT-PCR and Western blot respectively. Results: The levo-Anlodipine can decrease the free calcium concentration of the anoxic myocardial cell markedly(359.06 +/- 75.00 nM) and increase the expression of Ca(2+) ATPase of cytomembrane and sarcoplasmic reticulum under the condition of anoxia. Conclusion: The levo-Anlodipine can enhance the resistance to calcium overload of anoxic myocardial cells through increasing the expression of Ca(2+) ATPase.
