A Simple And Rapid Method For Simultaneous Determination Of Albiflorin And Paeoniflorin In White Peony Root

A simple and rapid high-performance liquid chromatographic (HPLC) method was developed and validated for the simultaneous determination of albiflorin and paeoniflorin in white peony root. The optimal conditions of separation and detection were achieved on a Thermo Hypersil ODS-2 Column (250 mmx4.6 mm, 5 mu m) by isocratic elution with the mobile phase of methanol-water (35:65). The detection wavelength was 230 nm and the column temperature was at 30 degrees C. The coefficients of linear regression of albiflorin and paeoniflorin were 0.9997 and 0.9992. The calibration curves showed that the linearity were completely acceptable. The average recovery (AR) of albiflorin and paeoniflorin was 98.36% and 96.99%, respectively. The RSD value of each analyte was less than 3.00%. It was an indication that the precision, repeatability, stability and accuracy were all in accordance to the principles. This method was successfully applied to determine 6 samples of white peony root collected from different areas and its ethanol extract. The results indicated that this method was simple, rapid and feasible for the quantification of albiflorin and paeoniflorin in white peony root.
