Stable isotope labeled peptide fragment synthesis and quantitation new method for quantitative proteomics research

The invention provides a universal, rapid and extensible way for generating labeled peptide fragment for quantitative determination. The method consists of: designing a peptide fragment as the following MGAGR-(XXX)5-25K/R-WSHPQFEK-GGD, integrating a to-be-synthesized internal standard peptide fragment (XXXK/R) with strep-tag (WSHPQFEK), using a cell-free protein synthesis system to synthesize the peptide fragment, and carrying out strep-tag affinity enrichment and complete enzymolysis so as to obtain a peptide fragment product synthesized by series connection of the stable isotope labeled peptide fragment and strep-tag in a molar ratio of 1:1. Precise quantitative analysis of the synthetic stable isotope labeled peptide fragment can be realized by quantitation of the series connection synthesized strep-tag.
