•YedK binds various DNA, particularly with higher affinity to those with single strand.•Short dsDNA can capture YedK, which is further enhanced by addition of ssDNA.•YedK represses plasmid transformation efficiency and copy number.•YedK represses plasmid transformation efficiency through the single strand binding activity.
