•S and N protein was simultaneously determined by the developed method.•The developed method was accurate and sensitive, and it was faster than vivo animal tests.•It can be used for quality evaluation of inactivated COVID-19 vaccine.•It can be used to evaluate other protein-based COVID-19 vaccines.
