•Rac2, Cdc42 and RhoH mutations resulted in immunological defects observed in PIDs.•Deregulation of RhoA, Rac1, RhoG and TC10 activities may also contribute to the phenotypes observed in PIDs.•RhoGEFs mutations may promote PID conditions by altering their associated small RhoGTPases activation status.
