Screening and identification of antisense accessible sites of survivin {\sl in vitro}

RT-PCR and Western blot analysis indicated that mRNA and protein levels of survivin in MKN-45 cells were all reduced significantly. Tetrazolium bromide (MTT) colorimetry demonstrated that cellular growth was sigificantly inhibited after transfection with 600 nmol/L AS-ODN_(1)--AS-ODN_(4) for 24 hours. Cellular apoptosis was detected by electronic microscopy, Annexin V-FITC and propidium iodide staining flow cytometry. The results indicated that AAS of survivin could be effectively selected in vitro by random oligonucleotide library/RNase H cleavage combining with computer software analysis, whose corresponding AS-ODNs could block the biological functions of survivin gene.
