The invention discloses a device and method for determining the positions of surface spots/blots of a transparent material. The device comprises a light source, a camera, a positioning device used forcamera positioning, a data processing device and a judging device, wherein the light source and the camera are positioned on the same side of the transparent material; natural light emitted by the light source is incident to the surface of the transparent material by using a brewster angle; the positioning device is used for positioning the camera onto a reflected light path after incident lightis incident through the upper surface or the lower surface of the transparent material; the camera is used for observing images on the upper surface and the lower surface respectively; the data processing device is used for recording incident position information and imaging information observed by the camera and processing the imaging information to obtain the strength of imaging; the judging device is used for judging the positions of the spots/blots according to the light intensity of the spots/blots observed by the camera. The device disclosed by the invention is simple in structure, and the judgment for the positions of the spots/blots of the transparent material can be realized accurately and quickly.
