•Major urinary metabolites of zinc pyrithione confirmed for the first time in vivo.•UHPLC-MS/HRMS assay enabled better selectivity, LOQs than conventional LC-MS/MS.•Simple “dilute and shoot” assay with short run time for clinical sample analysis.•Commercial urine lots contained trace levels of pyrithione metabolites.
