•BTZ injection the night before blood collection is effective for HSPC mobilization.•BTZ injection 3 days before blood collection adversely abrogated G-CSF-induced HSPC mobilization.•BTZ did not affect VLA-4 expression on HSPCs and VCAM-1 expression on BMSCs.•BTZ lowered CXCL12 chemokine expression in the bone marrow.•BTZ increased vascular permeability to further enhance HSPC mobilization.
