Understanding the effect of increased cell specific productivity on galactosylation of monoclonal antibodies produced using Chinese hamster ovary cells

•Consistent inverse correlation was observed between qP and %Gal across multiple cell lines, mAbs and cell culture processes.•Conditions that impacted mAb qP also impacted gene expression profiles of the N-glycosylation pathway.•For the high qP and low %Gal conditions, some or all the following genes B4GalT1, slc35a3 and slc39a8 were downregulated compared to control.•No significant impact on gene expression profiles in the N-glycosylation pathway was observed under the high pH condition (low qP, high %Gal).•Regulatory mechanisms that impact qP and %Gal are complex and strategies to modulate them are highly dependent on treatment conditions.
