•GPR10 deletion activates PrRP signaling in the NTS regulating energy balance.•Mild obesity with increased adipose tissue and liver weight in GPR10 KO males.•Lipogenesis genes in adipose tissue and liver are downregulated in GPR10 KO males.•Metabolic lipolytic genes are upregulated in GPR10 KO females.•Compensatory expression prevents adipose tissue and liver weight gain in females.
