Assessment of solid phase microextraction as a sample preparation tool for untargeted analysis of brain tissue using liquid chromatography-mass spectrometry

•Bio SPME and LC-HRMS was evaluated as an approach for untargeted brain analysis.•The effects of various parameters on the metabolites extracted were evaluated.•C18, MM, and HLB coatings were able to provide comparable brain tissue information.•SPME enabled the detection of ~800 (+ mode) and ~1200 (- mode) metabolite features.•Most model metabolites showed absolute matrix effects in the range of 80-120%.
