Construction Of 3d Electrochemical Cytosensor By Layer-By-Layer Assembly For Ultra-Sensitive Detection Of Cancer Cells

In the past 50 years, cancer has been the leading cause of death globally, and the development of effective cancer early diagnosis technology is still facing challenges. Herein, a three-dimensional (3D) mesoporous-macroporous interface for electrochemical sensing was constructed on the electrode surface through the layer-by-layer assembly method. Mesoporous silica (MS) coating was epitaxially grown on the outer side of the polystyrene (PS) microtubes. Then after loading the active component sambucus nigra agglutinin (SNA), the 3D mesoporous-macroporous interface was applied to the super-sensitive electrochemical detection of cancer cells. Specifically, the layer-by-layer assembly method was conducive to maximize the exposure of the detection of active substances, which had better cellular capture performance than direct mixing methods. The 3D structure of PS nanotubes was conducive to improving the specific surface area of the electrode, promoting its capture efficiency of cancer cells. The mesoporous structure can load more SNA, improving the specific recognition performance of cancer cells. The results showed that the detection platform based on SNA-SA specific recognition technology developed in this work had excellent cancer cell detection performance, stability, and selectivity, which is expected to be used in the early detection of cancer.
