•A 2D-LC-UV detection method for simultaneous determination of Asp, Glu, Gly, Tau and GABA was developed.•The 2D-LC-UV method has high loading capacity, excellent resolution, and good peak shape.•Neurotransmitters in blood and brain of rats and pigs were determined.
