Rutin Ameliorates Cardiac Dysfunction of Diabetic Rats

To investigate the effect of rutin on cardiac dysfunction of the diabetic rats and the relevant mechanism. The type 2 diabetic rats was established by injecting streptozoticin (STZ) combined with high-fat food. The rats of the rutin-treatment group were orally administrated with rutin at a dose of 200 mg/kg. day. The cardiac function was evaluated by serial echocardiography. The myocardial tissue morphology of rats was evaluated by hematoxylin-eosin (HE) staining and electron microscope. The expression of sarcoplasmic reticulum Ca2+-ATPase (SERCA), phospholamban (PLB) and ryanodine receptor (RyR2) were detected by immunofluorescence and western blot. The diabetic rats presented a disturbance cardiac function which were attenuated by the rutin treatment. Moreover, rutin can also increase the expression of SERCA and RyR2, and decrease the PLB level of the diabetic rats. Taken together, these results suggested that rutin can improve the cardiac dysfunction of the diabetic rats through regulating the calcium regulates protein expression.
