Quantification of Protodioscin in Tribulus terretris. Spray Dried Extract by RP-HPLC Diode Array Detection

Tribulus terrestris is an annual herb of Zygophyllaceae commonly known as Gohkru. It is widely used in traditional and folk medicine being rich source of steroidal saponins. Steroidal saponine protodioscin is the active biomarker for quality control of this plant extracts. The aim of current study was to quantify the steroidal saponine named protodioscin of T. terrestris spray dried extract by reverse phase HPLC method coupled with PDA detector. The solvent system was comprised of water and acetonitrile. A gradient elution was used for separation of biomarker at 200 nm and at flow rate of 1.0 mL/min. Separation was performed using C18 column and temperature of 40 degrees C. The method has been validated as per ICH guidelines for precision, accuracy, recovery and reproducibility, ruggedness and robustness by guidelines CPMP/ICH/381/95. The method was found precise, accurate, repealable, robust and reproducible according to guidelines of United Slates Pharmacopeia (LISP) 2014. The content of protodioscin was found to be 1.06 % as compare to standard. Use of spray drying technology can give multiple advantages in both food and pharmaceutical industry. As there is insufficient work reported on spray dried extract of T. terrestris so this study was established.
