•The validated HPLC method was reliable to measure IR780 drug loading in formulation.•The validated LC–MS/MS method was reliable to measure IR780 in mouse liver and lung.•Oral administered IR780 granules resulted in high concentration in mouse lungs at 2 h of post dosing.
