•The value of PowerPlex®18D used for forensic purpose in Guizhou Miao was evaluated.•Comprehensive population genetic comparisons of 174 Asian populations were conducted.•The genetic affiliations varied among Guizhou Miao tribes and other nationalities.•The possible routes of southward migration for Miao nationality were suggested.
