•Transcriptomic responses to polymyxin/chloramphenicol combination were deciphered using a systems study approach.•Emergence of polymyxin resistance was inhibited by chloramphenicol in NDM-producing Klebsiella pneumoniae.•Inhibition of cell metabolism was mainly driven by chloramphenicol.
