•Feather cleaning with medical gauze could reduce feather amount and simplify operation.•Hair filtration with medical gauze can overcome difficulty of feather centrifugation.•The extract was purified by MCX@ Fe3O4 nanoparticles.•Illegal use of azithromycin in broiler feather samples was confirmed by LC-MS/MS.
