•Lentivirus preparations can contain high and variable cytotoxicity.•To assess such cytotoxicity, a cell line-based sensitive bioassay is developed.•Unique workflows are defined that yield lentiviruses with nominal cytotoxicity.•These high-titer, low-cytotoxicity viruses efficiently transduce PB-HSPCs.•HSPC transduction can be boosted by cyclosporin H without cytotoxicity
