•Rapastinel is a positive NMDAR modulator with a novel mechanism of action.•Rapastinel produces rapid-acting and long-lasting antidepressant effects in rodents.•Rapastinel induces fewer CNS-related adverse side effects than ketamine in rodents.•Rapastinel does not produce locomotor or sensorimotor gating side effects in mice.•Rapastinel does not affect sleep architecture or increase gamma frequency in rats.
