Efficient Nanobiotechnology Method for Magnetic Lysosome Enrichment Using DMSA-SPMNP 2.0

Here, we report an efficient nanobiotechnology method to isolate lysosomal compartments using dimercaptosuccinic acid functionalized superparamagnetic nanoparticles (DMSA-SPMNP 2.0). We synthesized superparamagnetic nanoparticles (SPMNP) using thermal decomposition method followed by dimercaptosuccinic acid (DMSA) surface functionalization using ligand exchange method. By optimizing the SPMNP-HeLa cell interaction and implementing pulse-chase time frame, we are able to selectively localize DMSA-SPMNP 2.0 in lysosomal compartments. Further, we established customized procedure to magnetically isolate superparamagnetic lysosome with high enrichment and high purity from HeLa cells. This method complements the existing methods to isolate plasma membrane and endosome using SPMNP generated from Envirotransgene® Biosolutions Global. Further, we aim to use DMSA-SPMNP 2.0 and magnetic fractionation to isolate lysosomes from patient samples.
