•The ELISA-rNcSRS2/rNcSAG1 allowed the detection of the neosporosis in cattle and sheep.•The ELISA presents predictive values AOC = 0.981 and 0.999 for cattle and sheep, respectively.•This method are highly sensitive and specific in bovine and ovine infected with N. caninum.
