•FMD clinical signs range from mild to severe or subclinical.•Pathogenesis of SAT2 viruses did not correlate with cell killing or viral fitness.•SAT2 viruses have a wide range of lability at low pH or high temperatures.•No correlation was found between biophysical stability and pathogenesis.•Effective control of SAT2 viruses needs to consider variance in biological phenotypes.
