•Hyaluronic acid (HA)-functionalized osteopontin (OPN)-loaded PLGA nanoparticles were fabricated.•BSA/OPN-NPs have good morphology, size, zeta potential, dispersion and biocompatibility.•Surface HA functionalization of NPs targets colitis cells for drug delivery.•BSA/OPN-NPs enhanced the intestinal barrier and alleviated inflammation.•BSA/OPN-NPs alleviated inflammatory bowel disease through the NFκB signaling pathway.
