•2D-LC is applied as an investigational tool to facilitate antibody charge variant characterization.•Direct and efficient bridging between size and charge variants is achieved using both 2D AEX-SEC and 2D CEX-SEC.•Aggregates elute late on AEX and CEX columns presumably due to secondary hydrophobic interaction.•First reported 2D SEC-IEC application for antibody characterization.
