•Most clinical labs measure vitamins A & E using liquid-liquid extraction and HPLC.•This approach is laborious, often requiring dedicated staff and instrumentation.•We describe a simple, rapid protocol for measurement of vitamins A and E.•Use of multiplexing SPLC-MS/MS consolidates tests and reduces analysis times.
