•Mus m 1.0102 lipocalin allergen has three conserved Cysteines.•Disulfide bridge contributes to resistance to thermal stress and ligand affinity.•Free Cys substitution favours reversible unfolding and prevents aggregation.•The free Cys-Ala substitution mutant preserves allergenicity comparable to the wildtype.•Free Cys-Ala mutant is promising for diagnostics and specific immunotherapy.
