•[D-Trp7]-[Leu11]-HK-1 and [D-Trp9]-[Leu11]-HK-1 attenuates HK-1-induced scratching.•[D-Trp9]-[Leu11]-HK-1 attenuates chloroquine- and histamine-induced scratching.•[D-Trp9]-[Leu11]-HK-1 extends the duration of the anti-pruritic effect.•[D-Trp9]-[Leu11]-HK-1 dose-dependently attenuates pruritogen-induced scratching.•Intranasal administration of [D-Trp9]-[Leu11]-HK-1 attenuates scratching behavior.
