•PYM and BAM were enriched by column-based D157 resins in high yields.•P(st-dvb) microsphere based MPLC was firstly used for preparation of PYM and BAM.•This method was developed with environmentally-friendly aqueous mobile phase.•Operational parameters of column-based D157 resins and MPLC were optimized.•This paper shows the highest purities and recoveries of PYM and BAM so far.
