•LPS-induced septic condition leads to decreased expression of junctional proteins resulting in apoptosis of PMVECs.•Under LPS treatment, the application of hydrogen-rich medium can inhibit the activity of RhoA and recover cell viability.•Hydrogen treatment can prevent LPS-induced endothelial barriers destruction through RhoA inhibition.
