•Coiled-coils serve as a platform for protein-protein interactions.•L. donovani coronin CC possesses an inherent asymmetry, caused by steric hindrance.•Residues mutated, biophysically and structurally characterized.•Two of the mutants have a lower Tm value.•One of the helix in two of the mutant structures show a ~2 turn translation.
