Proteomics insights into the effects of MSTN on muscle glucose and lipid metabolism in genetically edited cattle.

•Comparative proteomic and phosphoproteomic analyses of gluteus muscle tissues from MSTN−/− transgenic cattle were performed to explore the signaling pathway of MSTN in metabolic cross-talk and cellular metabolism.•A total of 72 differentially expressed proteins (DEPs) and 36 differentially expressed phosphoproteins (DEPPs) were identified in MSTN−/− cattle compared to WT cattle based on the TMT6-plex labeling method.•The DEPs were categorized as muscle, translation and lipid metabolism related proteins along with mitochondrial activity associated proteins, while the DEPPs were mainly associated with glycol-metabolism pathway.•The fatty acid β-oxidation process and glycolysis pathway were increased and the content of intramuscular fat and glycogen were decreased in MSTN−/− cattle.•MSTN knockout triggers the activation of AMPK signaling pathways to regulate glucose and lipid metabolism by increasing the AMP/ATP ratio.
