•The protein identification analysis identified five differentially expressed proteins in the OTA-GES-1T cells group.•The expressions of Annexin A3 both at protein and mRNA levels increased in OTA-GES-1T cells group.•Annexin A3 siRNA transfection could inhibit the proliferation of OTA-GES-1T cells.•Annexin A3 siRNA transfection could lead cells to be arrested at G0/G1 phase in OTA-GES-1T cells group.•Inhibition Annexin A3 by siRNA effectively prevented the migration and invasion abilities of OTA-GES-1T cells.
