Separating distinct structures of multiple macromolecular assemblies from cryo-EM projections

•Single particle cryo-EM can be performed on mixtures containing multiple distinct macromolecular assemblies.•Developed an algorithm using common lines in a graphical framework to classify 2D projection images.•Using this algorithm, we separate a mixture of synthetic data and a mixture of experimental data.
