•The protease ZMPSTE24 performs the final cleavage step in prelamin A maturation.•Mutations in ZMPSTE24 or LMNA cause progeria and related disorders.•A “humanized yeast system” to examine ZMPSTE24 and LMNA variants was developed.•This system can analyze cleavage activity for ZMPSTE24 and LMNA disease alleles.
