Introduction: Changes in the ratio of reduced (GSH) to oxidized (GSSG) glutathione is commonly used to indicate redox status in the heart. However, previously reported values of myocardial GSH, GSSG and redox state (GSH:GSSG) are conflicting, possibly due to a lack of sensitive, accurate quantification of GSH and GSSG, as well as blood contamination and auto-oxidation during sample preparation. LC-MS/MS is a gold standard technique for the quantification of low molecular compounds but not commonly used for redox state assessment in heart tissue. Fatty acids (palmitate) are an important fuel for the heart and are suggested to influence cardiac glutathione homeostasis. Hypothesis: Elevated palmitate levels will influence cardiac redox state (GSH:GSSG) following oxidative stress induced by diamide exposure or ischemia/reperfusion (I/R).
