•We developed a green methodology to reduce ethyl acetoacetate by immobilized cells.•A new solvent system formed by supercritical carbon dioxide and glycerol was proposed.•Operational stability and viability of the biocatalyst was also evaluated.•Proposed methodology successfully reduced the substrate to an enantiopure alcohol.•This approach could be used to optimize large scales production of secondary alcohols.
